The Microtox Chronic Toxicity Test (Chronic) is a fast chronic bioassay based on bacterial luminescence which detects and quantifies chronic toxicity. The luminescent microorganisms, Vibrio fischerii, emit light as a normal consequence of respiration. Chemicals or chemical mixtures which are toxic to the bacteria cause a reduction in light output proportional to the toxicity. Many different sample types and phases can be tested with the Chronic test.
The system is two-part, consisting of the microorganism used in the bioassay, and a temperature-controlled luminometer which reads the bacterial light output. A computer is linked to the system to provide rapid processing of the data.
The key to making system replicable and uniform is the "reagent," freeze-dried microorganisms. Lyophilized under strict controls at Microbics Corporation, the bacteria are exceptionally consistent in their response to toxic materials. Additionally, the freeze-drying process allows for storage of the reagent for up to 1 year in a standard deep freezer (-20C) before use. There are no stock cultures to maintain, no special environmental chambers to construct. Simply reconstitute the lyphilized bacteria, and then use them in the Microtox Chronic Test.
Volumes are kept small in the Microtox Chronic test. Only a few milliliters of sample are necessary to run the test. This makes sample collection, storage, and disposal easy and inexpensive. The test is easy to perform, taking only basic lab skills. It yields NOEC/LOEC endpoints, as well as ICXX on samples.

AZUR Environmental is a world leader in bioluminescence technology. The Microtox Chronic Toxicity Test is one of three bioassays which have been developed at Microbics. The other two are the Microtox Acute Toxicity Test, and the Mutatox test, which detects genotoxicity and mutagenicity. Microtox Chronic is a logical successor to Microtox Acute where extreme sensitivity is necessary. It is as much as 100 times more sensitive than the Acute test, and its sensitivity is equivalent to that of the most sensitive regulatory chronic bioassays now in use.
Limitations:
All bioassays, including Microtox Chronic, have the same essential limitation. They measure toxicity, and can only determine whether a sample is toxic or not, rather than indicating what specific chemicals are present. Chemical detection methods like GC/MS, AA, and HPLC are often used in conjunction with bioassays for this reason.
